Target data

Function

Generates phosphatidylinositol 3-phosphate (PtdIns3P) and phosphatidylinositol 3,4-bisphosphate (PtdIns(3,4)P2) that act as second messengers. Has a role in several intracellular trafficking events. Functions in insulin signaling and secretion. Required for translocation of the glucose transporter SLC2A4/GLUT4 to the plasma membrane and glucose uptake in response to insulin-mediated RHOQ activation. Regulates insulin secretion through two different mechanisms: involved in glucose-induced insulin secretion downstream of insulin receptor in a pathway that involves AKT1 activation and TBC1D4/AS160 phosphorylation, and participates in the late step of insulin granule exocytosis probably in insulin granule fusion. Synthesizes PtdIns3P in response to insulin signaling. Functions in clathrin-coated endocytic vesicle formation and distribution. Regulates dynamin-independent endocytosis, probably by recruiting EEA1 to internalizing vesicles. In neurosecretory cells synthesizes PtdIns3P on large dense core vesicles. Participates in calcium induced contraction of vascular smooth muscle by regulating myosin light chain (MLC) phosphorylation through a mechanism involving Rho kinase-dependent phosphorylation of the MLCP-regulatory subunit MYPT1. May play a role in the EGF signaling cascade. May be involved in mitosis and UV-induced damage response. Required for maintenance of normal renal structure and function by supporting normal podocyte function. Involved in the regulation of ciliogenesis and trafficking of ciliary components (By similarity).

Notes

ab307099 is the carrier-free version of Anti-PI3-Kinase p170 antibody [17/PI3-Kinase p170] ab307098.

This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. The carrier-free buffer and high concentration allow for increased conjugation efficiency.

This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Supplementary info

Activity summary

PI 3 Kinase Class 2A also known as Cpk is a significant enzyme involved in the regulation of various cellular processes. The target weighing approximately 170 kDa acts as a lipid kinase that phosphorylates phosphatidylinositol converting PI to PI3P. Cpk expresses mainly in tissues like the liver muscle and heart where it contributes to cellular homeostasis and metabolic regulation.

Biological function summary

This enzyme family modulates intracellular signaling cascades influencing processes such as cell growth survival and metabolism. Cpk forms part of a larger complex that interacts with adaptor proteins facilitating its recruitment to cellular membranes. This interaction is important for its signaling activity and function in maintaining cellular equilibrium.

Pathways

PI 3 Kinase Class 2A plays an integral role in the insulin signaling pathway and the TOR signaling pathway. In the insulin pathway it collaborates with proteins like Akt to modulate glucose uptake and storage in cells. Similarly in the TOR pathway Cpk contributes to nutrient sensing and cell growth regulation interconnected with proteins such as mTOR affecting protein synthesis and autophagy.

Associated diseases and disorders

Dysregulation of PI 3 Kinase Class 2A links to cancer and type 2 diabetes. Alterations in its activity may lead to abnormal cell proliferation and survival characteristics of cancerous transformations associated with proteins like PTEN. Meanwhile its role in glucose metabolism ties it to insulin resistance and diabetes pathogenesis where interactions with insulin receptor substrates become important for disease progression.
