Supplementary information

PI4 kinase beta also known as PI4KB is an important enzyme involved in lipid signaling. It catalyzes the phosphorylation of phosphatidylinositol (PI) to produce phosphatidylinositol 4-phosphate (PI4P) an essential lipid for various cellular processes. PI4KB has a molecular mass of approximately 92 kDa. This enzyme shows expression in various tissues including the brain liver and pancreas. In cells PI4KB predominantly localizes at the Golgi apparatus and some plasma membrane sites.
Biological function summary

The enzyme plays a role in maintaining proper intracellular membrane structure and function. It operates as part of a larger complex involving additional elements such as the small GTPase ARF1 and the scaffold protein ACBD3 which are necessary for its recruitment and activity at Golgi membranes. PI4KB facilitates vesicular trafficking and supplies substrates for the production of other phosphoinositides impacting diverse cellular events like cell growth and signal transduction.

Pathways

PI4KB is an important participant in the phosphoinositide signaling pathway. This pathway regulates membrane dynamics and trafficking influencing cell signaling and membrane identity. PI4KB interacts with proteins such as PI4K2A and PI4K2B in the synthesis of different phosphoinositides which are components of the signaling cascade that include PI3K/AKT pathway having implications in cell growth and survival.

PI4KB has associations with certain infectious diseases and neurological disorders. The enzyme is linked to hepatitis C virus (HCV) replication where it interacts with viral proteins like NS5A facilitating the formation of replication complexes. Furthermore PI4KB has links to neurodegenerative disorders like Alzheimer's disease possibly through its influence on vesicular trafficking and lipid metabolism. These associations highlight its importance in disease mechanisms and as a potential therapeutic target.

Target data

Phosphorylates phosphatidylinositol (PI) in the first committed step in the production of the second messenger inositol-1,4,5,-trisphosphate (PIP). May regulate Golgi disintegration/reorganization during mitosis, possibly via its phosphorylation. Involved in Golgi-to-plasma membrane trafficking (By similarity) (PubMed : 10559940, PubMed : 11277933, PubMed : 12749687, PubMed : 9405935). May play an important role in the inner ear development.. (Microbial infection) Plays an essential role in Aichi virus RNA replication (PubMed : 22124328, PubMed : 22258260, PubMed : 27989622). Recruited by ACBD3 at the viral replication sites (PubMed : 22124328, PubMed : 27989622).. (Microbial infection) Required for cellular spike-mediated entry of human coronavirus SARS-CoV.
