Pore-forming subunit of the mechanosensitive non-specific cation Piezo channel required for rapidly adapting mechanically activated (MA) currents and has a key role in sensing touch and tactile pain. Piezo channels are homotrimeric three-blade propeller-shaped structure that utilize a cap-motion and plug-and-latch mechanism to gate their ion-conducting pathways. In inner ear hair cells, PIEZO1/2 subunits may constitute part of the mechanotransducer (MET) non-selective cation channel complex where they may act as pore-forming ion-conducting component in the complex. Required for Merkel-cell mechanotransduction. Plays a major role in light-touch mechanosensation.

PIEZO2 also known as FAM38B is a mechanosensitive ion channel protein. It has a high molecular mass of approximately 287 kDa. This protein exists in mechanosensitive tissues such as the skin bladder and lungs. PIEZO2 detects mechanical stimuli by opening its ion-conducting pore allowing the influx of cations into the cell which sends a signal.
PIEZO2 plays an important role in the sensory pathways of touch and proprioception. It is not a part of a larger protein complex but operates as a homotrimer. The channel's sensitivity to mechanical forces enables cells to transduce physical touch into electrical signals facilitating an organism's ability to perceive its environment.
Sensory transduction and mechanotransduction heavily involve PIEZO2. This protein interacts within pathways that transmit information about mechanical loads to the nervous system. Although not directly part of larger pathways' cascades its role is well observed in touch-related signaling paths where it occasionally interacts with proteins like TRPV1.
Mutations in PIEZO2 relate to Gordon Syndrome and Marden-Walker Syndrome. Dysfunction in PIEZO2 can lead to defects in touch and balance implicating disorders with sensory processing. There is also a notable connection to the protein TRPA1 which participates in similar sensory pathways and can correlate with sensory-related dysfunctions.
