PIF1 also known as Pif1p is a helicase enzyme that plays a critical role in DNA replication and repair. It is part of the PIF1 family of helicases which are conserved from bacteria to humans. The enzyme has a molecular mass of approximately 70 kDa. PIF1 locates itself in the mitochondria and the nucleus where it acts on both nuclear and mitochondrial DNA. The enzyme functions by unwinding DNA helices facilitating various genetic processes such as replication recombination and repair.
Biological function summary

The protein maintains genome stability by resolving R-loops and DNA structures that can hinder replication. PIF1 interacts with proteins involved in DNA maintenance but it is not typically known to be part of a larger protein complex. The helicase is noted for suppressing the instability of genomic regions with G-quadruplex structures being particularly active at telomeres. Its activity prevents the accumulation of deleterious DNA structures that might otherwise disrupt genomic integrity.

Pathways

PIF1 engages in DNA replication and repair pathways. Within these biological pathways PIF1 ensures correct fork progression and prevents stalling which is linked to several other proteins such as Replication Protein A (RPA) and DNA pol Δ. PIF1 also has a role in the maintenance of telomeres interacting with telomeric proteins to prevent aberrant telomere elongation and reduce genomic instability.

PIF1's function relates closely to certain cancer types and mitochondrial disorders. Deficiencies or mutations in the PIF1 helicase can result in genomic instability contributing to tumor progression. PIF1 has been linked with BRCA1 in the context of breast cancer where it may play a complementary or compensatory role. Additionally defects in mitochondrial PIF1 functionality can contribute to mitochondrial myopathies disorders that involve proteins like mitochondrial DNA polymerase γ.
