The phosphatidylinositol glycan class A protein (PIGA) plays an important role in the biosynthesis of glycosylphosphatidylinositol (GPI) anchors which tether various proteins to the cell membrane. The PIGA protein with a molecular weight of approximately 50 kDa is expressed predominantly in hematopoietic cells and other tissues with high protein turnover. It functions as a catalytic subunit in the synthesis of GPI anchors an important class of glycolipids facilitating the attachment of proteins to the cell surface.
Biological function summary

PIGA facilitates the early steps of GPI anchor formation where it acts as part of a multi-protein complex including PIGC PIGH and PIGP. This complex catalyzes the transfer of N-acetylglucosamine to phosphatidylinositol initiating GPI anchor assembly. The absence or defect of PIGA disrupts GPI anchor biosynthesis affecting the delivery and function of GPI-anchored proteins important for cell signaling cell adhesion and enzyme functions.

Pathways

PIGA's role is important in the early stages of the GPI biosynthetic pathway interconnected with the ER-associated degradation pathway (ERAD). Within these pathways PIGA interacts with proteins such as PIGM and PIGX orchestrating sequential enzymatic reactions and ensuring successful GPI anchor synthesis. These pathways later impact various cellular processes including protein traffic and membrane organization.

Mutations or deficiencies in PIGA lead to conditions like paroxysmal nocturnal hemoglobinuria (PNH) and certain forms of inherited GPI deficiency (IGD) syndromes. In PNH defective GPI anchor biosynthesis causes red blood cells to lack protective CD55 and CD59 proteins resulting in increased hemolysis. Additionally similarities in GPI biosynthesis dysfunctions link PIGA to PNH-related hemolytic disorder and neurological impairments in IGD syndromes involving associated proteins like PIGM and PIGN.
