The phosphatidylinositol-4-phosphate 5-kinase type 1 beta (PIP5K1β) also known as PIPK type I beta is an enzyme that plays an important role in the synthesis of phosphatidylinositol 45-bisphosphate (PIP2) from phosphatidylinositol 4-phosphate (PI4P). This enzyme has a molecular weight of approximately 68 kDa. It expresses across various tissues with significant presence in the brain and in hematopoietic cells. The PIP5K1β enzyme is important in regulating the levels of PIP2 a critical lipid in membrane dynamics and signaling.
Biological function summary

PIP5K1β acts within the phosphoinositide signaling pathway which is essential for a range of cellular processes including actin cytoskeleton organization vesicle trafficking and cell survival. It forms part of a complex with other kinases and phosphatases that modulate cellular lipid concentrations. PIP5K1β also contributes to signal transduction processes by participating in the recruitment and activation of proteins that respond to membrane-associated signals.

Pathways

PIP5K1β is heavily involved in the regulation of the phosphoinositide 3-kinase (PI3K) pathway and the actin cytoskeleton pathway. Its product PIP2 serves as a substrate for PI3K which generates phosphatidylinositol-345-trisphosphate (PIP3) an important signaling molecule. PIP5K1β therefore indirectly impacts pathways that involve proteins like AKT and Rac1 which are important to cellular growth and motility.

PIP5K1β connects to cancer and immune system-related disorders. Dysregulation of PIP5K1β activity can lead to aberrant PI3K signaling contributing to oncogenic processes. Additionally it affects proteins such as AKT which plays a role in tumor progression and survival. In immune disorders altered PIP5K1β levels can impact the actin cytoskeleton affecting immune cell dynamics and function.

Catalyzes the phosphorylation of phosphatidylinositol 4-phosphate (PtdIns(4)P/PI4P) to form phosphatidylinositol 4,5-bisphosphate (PtdIns(4,5)P2/PIP2), a lipid second messenger that regulates several cellular processes such as signal transduction, vesicle trafficking, actin cytoskeleton dynamics, cell adhesion, and cell motility (By similarity). PtdIns(4,5)P2 can directly act as a second messenger or can be utilized as a precursor to generate other second messengers : inositol 1,4,5-trisphosphate (IP3), diacylglycerol (DAG) or phosphatidylinositol-3,4,5-trisphosphate (PtdIns(3,4,5)P3/PIP3) (By similarity). Mediates RAC1-dependent reorganization of actin filaments. Contributes to the activation of phospholipase PLD2. Together with PIP5K1A, is required, after stimulation by G-protein coupled receptors, for the synthesis of IP3 that will induce stable platelet adhesion (By similarity).
