PIP5K2 gamma also known as phosphatidylinositol-5-phosphate 4-kinase type-2 gamma is an enzyme encoded by the PIP4K2C gene. It plays an important role in cellular processes by phosphorylating phosphatidylinositol-5-phosphate to generate phosphatidylinositol-45-bisphosphate. The molecular mass of PIP5K2 gamma is approximately 51 kDa. This protein is mostly expressed in brain tissue but researchers have also found it in other tissues including the pancreas and spleen.
Biological function summary

The protein's primary role involves the regulation of phosphoinositide signaling pathways. PIP5K2 gamma participates in membrane trafficking cytoskeletal organization and signal transduction processes. It associates with different protein complexes that modulate these cellular activities. By controlling the levels of phosphatidylinositol-45-bisphosphate PIP5K2 gamma indirectly regulates several downstream signaling events.

Scientists have shown that PIP5K2 gamma is a part of the phosphatidylinositol signaling system which is important for signal transduction and cellular communication. It interacts closely with proteins such as phospholipase C and AKT. PIP5K2 gamma also contributes to the insulin signaling pathway impacting glucose metabolism and cell proliferation.

Studies link PIP5K2 gamma to several medical conditions. Its dysregulation associates with neurological disorders including bipolar disorder. Variations in this protein are also connected to cancer. For example in cancer alterations in the PIP5K2 gamma activity may affect pathways involving the AKT protein leading to uncontrolled cell growth and survival.

Phosphatidylinositol 5-phosphate 4-kinase with low enzymatic activity. May be a GTP sensor, has higher GTP-dependent kinase activity than ATP-dependent kinase activity. PIP4Ks negatively regulate insulin signaling through a catalytic-independent mechanism. They interact with PIP5Ks and suppress PIP5K-mediated PtdIns(4,5)P2 synthesis and insulin-dependent conversion to PtdIns(3,4,5)P3 (PubMed : 31091439).
