Supplementary information

Pirin also known as PIR protein is a nuclear iron-dependent non-heme oxidoreductase involved in various cellular processes. The PIR protein sometimes called PIR-B or PIR-A/B has a molecular mass of approximately 32 kDa. It expresses in many human tissues including the brain heart and lungs. The PIR protein plays significant roles in stress response and regulation of transcription factors making it a subject of interest in cellular regulation and signal transduction research.
Biological function summary

PIR protein associates with transcriptional co-factors and mediates gene expression regulation impacting cellular metabolism and growth. It interacts with several alternative forms such as PIR alternative and antipirin contributing to its diverse functionality. PIR often forms part of transcription complexes facilitating the modulation of gene networks essential for maintaining cellular homeostasis.

Pathways

PIR protein participates in significant biological processes like the NF-kB signaling and apoptosis regulatory pathways. In the NF-kB pathway PIR contributes to transcription factor activation while in apoptosis it influences pathways promoting cell survival or programmed cell death. PIR protein relates closely to proteins such as Bcl-3 and related transcription factors shaping cellular responses to external stimuli and maintaining balance between cell survival and death.

PIR protein associates with various cancers and neurodegenerative diseases. In cancer PIR's role in transcription regulation impacts tumor progression and metastasis interacting with proteins like c-Myc. In neurodegeneration abnormalities in PIR expression or function link to disorders such as Alzheimer's disease where it interacts with tau protein exacerbating the disease pathology. Understanding PIR's relationship to these conditions might provide insights into new therapeutic approaches.

Target data

Transcriptional coregulator of NF-kappa-B which facilitates binding of NF-kappa-B proteins to target kappa-B genes in a redox-state-dependent manner. May be required for efficient terminal myeloid maturation of hematopoietic cells. Has quercetin 2,3-dioxygenase activity (in vitro).
