Supplementary information

PKE also known as Protein Kinase Epsilon is a serine/threonine kinase with a mass of approximately 82 kDa. It belongs to the protein kinase C (PKC) family specifically the epsilon isoform. PKE expresses in a variety of tissues including brain heart and lungs where it plays a role in signal transduction processes. It achieves these functions through phosphorylation of target proteins modifying their activity localization and overall function.
Biological function summary

PKE influences cellular signaling and regulates important aspects of cell survival proliferation and differentiation. It associates with other proteins to form complexes that drive intracellular activities. PKE also takes part in neuron-related processes contributing significantly to maintenance of cellular integrity. Its activity affects cellular responses to extrinsic stimuli regulating pathways affecting diverse cellular outcomes.

Pathways

PKE integrates within signaling cascades such as the MAPK/ERK and NF-kB pathways. These pathways influence important cellular processes including growth and inflammation. In these systems PKE interacts with proteins like RAF1 and IKBKB driving downstream effects that alter gene expression and cellular responses. This involvement in such pathways highlights PKE's role in orchestrating complex biological processes.

PKE has implications in cancer and cardiac disorders. In cancer PKE's altered expression or activity impacts cell proliferation and survival interacting with other proteins such as RAS and AKT to promote oncogenic signaling. In cardiac disorders PKE modulates heart function and response to stress often implicating proteins such as TNNT2. Understanding PKE's role in these conditions aids in the development of targeted therapies that modulate its activity for therapeutic gain.
