Plasmin dissolves the fibrin of blood clots and acts as a proteolytic factor in a variety of other processes including embryonic development, tissue remodeling, tumor invasion, and inflammation. In ovulation, weakens the walls of the Graafian follicle. It activates the urokinase-type plasminogen activator, collagenases and several complement zymogens, such as C1 and C5. Cleavage of fibronectin and laminin leads to cell detachment and apoptosis. Also cleaves fibrin, thrombospondin and von Willebrand factor. Its role in tissue remodeling and tumor invasion may be modulated by CSPG4. Binds to cells (By similarity).

Angiostatin a fragment derived from plasminogen acts critically in inhibiting angiogenesis. It has a molecular mass of approximately 38 kDa. Angiostatin primarily derives from proteolytic cleavage of plasminogen within the vascular system. Expression of angiostatin occurs in various tissues but it is predominantly observed around tumor sites where it exerts its anti-angiogenic effects.
Angiostatin functions to inhibit the proliferation and migration of endothelial cells thereby blocking new blood vessel formation. It achieves this by binding to ATP synthase on the surface of endothelial cells. This protein operates as an isolated entity and does not typically form part of a larger complex. Angiostatin’s activity is essential in controlling physiological processes requiring reduced blood vessel development.
Angiostatin participates actively in the angiogenesis pathway interacting with factors like vascular endothelial growth factor (VEGF). It opposes pro-angiogenic signals by suppressing these pathways reducing the available nutrients to target tissues such as tumors. Angiostatin's role in the lysine-binding pathway also connects it to proteins like plasmin influencing fibrinolytic activity.
Angiostatin associates strongly with cancer and ocular diseases like diabetic retinopathy. Its inhibitory effect on angiogenesis restricts the blood supply to tumors aiding in cancer treatment. In cancer angiostatin interacts with plasminogen activator inhibitor-1 (PAI-1) to modulate plasminogen activity. In ocular diseases angiostatin’s impact on abnormal blood vessel growth contributes to therapeutic strategies to manage these conditions.
