Flow Cytometry (Intracellular) - Anti-Plastin L antibody [EPR4278] - BSA and Azide free (AB247773)

This data was developed using ab109129, the same antibody clone in a different buffer formulation.

Flow Cytometry analysis of MDA-MB-435S (Human mammary gland ductal carcinoma melanocyte) cells labeling Plastin L with purified ab109129 at 1/1000 dilution (1 μg/mL) (Red). Cells were fixed with 4% Paraformaldehyde and permeabilised with 90% Methanol. A Goat anti rabbit IgG (Alexa Fluor® 488, ab150077) secondary antibody was used at 1/2000. Isotype control - Rabbit monoclonal IgG (Black). Unlabeled control - Cell without incubation with primary antibody and secondary antibody (Blue).

What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Supplementary information

Plastin L also known as L-Plastin or plastine is a type of actin-bundling protein involved in regulating cytoskeletal rearrangements. This protein has a mass of approximately 70 kDa and serves an essential role in maintaining cell structure and mobility by cross-linking actin filaments. It is predominantly found in hematopoietic cells such as leukocytes but also expressed in certain non-hematopoietic cells. In zebrafish studies plastin L has been important for understanding its conservation and function across species.
Biological function summary

Plastin L participates in cellular processes such as phagocytosis cell motility and immune responses by stabilizing actin filaments. It does not form part of a larger protein complex but interacts with actin to perform its functions. As an actin-binding protein it helps in the formation and maintenance of the cytoskeleton impacting the cell's mechanical properties and response to external stimuli.

Pathways

Plastin L plays significant roles in immune cell signaling and cytoskeletal rearrangements. It is involved in pathways such as the immune response and cell adhesion where it contributes to leukocyte emigration and inflammation. Plastin L is functionally related to proteins like talin and paxillin which are key in mediating signal transduction resulting in cytoskeletal remodeling.

Plastin L is associated with cancer and immune system disorders. Overexpression of plastin L correlates with various forms of cancer highlighting its role in tumor cell invasion and metastasis. Additionally it shows a connection with autoimmune conditions where deregulation of actin polymerization affects immune cell function. In these contexts proteins like cofilin interact with plastin L to modulate actin dynamics and cellular responses.
