Immunocytochemistry/ Immunofluorescence - Anti-PLCG1 (phospho Y783) antibody [EPR1898Y] - BSA and Azide free (AB326611)
This data was developed using ab76031, the same antibody clone in a different buffer formulation.
Immunocytochemistry/Immunofluorescence analysis of Jurkat +/- pervanadate (1mM 30min) and Jurkat + pervanadate (1mM 30min) + LP labelling Phospholipase C gamma 1/PLC-gamma-1 (phospho Y783) with ab76031 at a dilution of 1/100. Cells were fixed with 4% paraformaldehyde and permeabilized with 0.1% tritonX-100. ab150077 (goat anti-rabbit IgG Alexa Fluor® 488) (1/1000) was used as the secondary antibody. The cells were co-stained with ab195889 (Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594)) at a 1/200 dilution. Nuclei counterstained with DAPI (blue). Control : PBS instead of the primary antibody.

Dot blot analysis of Phospholipase C gamma 1/PLC-gamma-1 (phospho Y783) phospho peptide (Lane 1) and Phospholipase C gamma 1/PLC-gamma-1 non-phospho peptide (Lane 2) labelling Phospholipase C gamma 1/PLC-gamma-1 (phospho Y783) with ab76031 at a dilution of 1/1000. A Goat Anti-Rabbit IgG (H+L) Peroxidase conjugated (ab97051) was used as the secondary antibody at a dilution of 1/20000. Blocking buffer : 5% NFDM/TBST. Dilution buffer : 5% NFDM /TBST.

Biological function summary
PLC-gamma-1 is integral in signal transduction and is an important player in the immune response. The protein does not function as part of a large multi-protein complex but interacts transiently with other signaling molecules. It acts downstream of growth factor receptors and immunoreceptors by mediating calcium release and activation of protein kinase C influencing cell proliferation migration and differentiation. Its expression pattern suggests a critical role in the nervous and immune systems affecting learning memory and immune defense.
Pathways
PLC-gamma-1 is an important component in both the phospholipase C and PLC-gamma pathways. These pathways include important interactions with proteins such as the receptor tyrosine kinases and Src family kinases. In these pathways PLC-gamma-1 modulates signals that guide cellular responses to external stimuli affecting processes like growth signaling and T-cell receptor activation which is vital in adaptive immunity.
