This data was developed using the same antibody in a different buffer formulation (ab32528).

ab32528 was shown to react with PLEC in wild-type U-2 OS cells in Western blot with loss of signal observed in a PLEC knockout cell line. Wild-type U-2 OS and PLEC knockout cell lysates were subjected to SDS-PAGE. Membranes were blocked in 5% milk in TBST for 1 hr before incubation with ab32528 overnight at 4 °C at a 1/5000 dilution. Blots were incubated with goat anti-rabbit HRP secondary antibodies at 0.2ug/mL before imaging. These data were provided by YCharOS Inc., an open science company with the mission of characterizing commercially available antibody reagents for all human proteins. Abcam and YCharOS are working together to help address the reproducibility crisis by enabling the life science community to better evaluate commercially available antibodies.

Plectin also known as PLEC is a large protein with a molecular mass of approximately 500 kDa. It serves as a cross-linking element that plays an important role in maintaining the structural integrity of various cells. Plectin is widely expressed in tissues such as muscle skin and nervous tissue. Its mechanical function involves linking intermediate filaments to membrane complexes and other cytoskeletal structures which supports cellular architecture and stability.
Biological function summary

Plectin contributes significantly to the structural organization of cells and tissues. It is a part of the cytoskeletal complex and assists in cellular integrity by interacting with actin microfilaments microtubules and intermediate filaments. Plectin provides elasticity to cells allowing them to withstand mechanical stress.
