Blocking and diluting buffer and concentration : 5% NFDM/TBST.
Low expression : P815.
The expression profile/ molecular weight observed is consistent with what has been described in the literature (PMID : 16715077, PMID : 12559962, PMID : 25485759, PMID : 35508542).
In Western blot, Anti-Vinculin antibody [EPR8185] (ab129002) staining at 1/10000 dilution.
This blot was developed using a high sensitivity ECL substrate. The high-sensitivity ECL substrate used allows for the detection of proteins in the mid-femtogram range.

This data was developed using ab321805, the same antibody clone in a different buffer formulation.
Blocking and diluting buffer and concentration : 5% NFDM/TBST.
Low expression : liver (PMID : 8806667, PMID : 12559962).
The expression profile/ molecular weight observed is consistent with what has been described in the literature (PMID : 16715077, PMID : 12559962, PMID : 25485759, PMID : 35508542).
To minimize protein degradation, tissues were lysed immediately after harvest and then applied to a gel and transfer membrane for Western blotting as soon as possible.
The identity of the lower MW band at approximately 37 kDa (in lane 2) is unknown.

Biological function summary
Plexin A1 functions in neural patterning and the immune response. It is a part of semaphorin signaling complexes that include neuropilins and other plexins. These complexes are critical in translating extracellular cues into intracellular responses impacting cellular dynamics such as motility and adhesion. Plexin A1 also interacts with various intracellular signaling molecules influencing actin cytoskeleton reorganization which is essential for proper neural development.
Pathways
Plexin A1 participates in semaphorin signaling and cytoskeletal dynamics pathways. It interacts with Sema3A and Sema4D proteins which are key signaling semaphorins. These interactions regulate cellular movement by influencing the activity of downstream effectors like Rho and Rac GTPases. The involvement of Plexin A1 in these pathways is important for processes like axon guidance and neuronal connectivity.
