Target data

Function

Receptor for SEMA5A that plays a role in axon guidance, invasive growth and cell migration. Stimulates neurite outgrowth and mediates Ca(2+)/Mg(2+)-dependent cell aggregation. In glioma cells, SEMA5A stimulation of PLXNB3 results in the disassembly of F-actin stress fibers, disruption of focal adhesions and cellular collapse as well as inhibition of cell migration and invasion through ARHGDIA-mediated inactivation of RAC1.

Activity summary

Plexin-B3 also known as PLXNB3 is a membrane protein with an estimated mass of approximately 215 kDa. It functions as a receptor for semaphorins particularly Sema5A in cell signaling processes. Plexin-B3 influences actin cytoskeleton rearrangement by interacting with the Rho GTPase family members. It is expressed in several tissues including the brain kidney and heart which suggests its diverse roles in different cellular contexts.

Biological function summary

Plexin-B3 modulates cellular processes such as growth adhesion migration and axon guidance. It forms complexes with other proteins such as Met and ErbB2 indicating its involvement in diverse signaling pathways. This protein plays an important role in the nervous system influencing neural circuit formation and neuronal connectivity. Its function in cell migration and adhesion also implicates it in cancer metastasis-related activities in various tumors.

Pathways

Plexin-B3 participates in the semaphorin signaling pathway which is important for guiding neuronal axons and other migratory cells. It engages with proteins like RhoA and Rac1 to transmit downstream signals that affect cytoskeletal dynamics. The role of Plexin-B3 in these pathways highlights its importance in maintaining cellular architecture and regulation of cell movement.

Associated diseases and disorders

Plexin-B3 has connections to certain cancers including prostate and brain cancer. Its interaction with Met a receptor tyrosine kinase links it to oncogenic processes including enhanced cellular proliferation and migration. Furthermore studies suggest a potential involvement in neurological disorders such as autism given its critical role in neural circuit formation and guidance although this requires further investigation.
