A549 (human lung carcinoma cell line) cells stained for PLS1 (green) using ab236976 at 1/133 dilution in ICC/IF, followed by Alexa Fluor 488® conjugated Goat Anti-Rabbit IgG (H+L). Counterstained wit DAPI.

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the antibody overnight at 4°C.

Paraffin-embedded human small intestine tissue stained for PLS1 using ab236976 at 1/400 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

PLS1 also known as Plastin-1 or fimbrin acts as a pivotal actin-bundling protein influencing the dynamic restructuring of the actin cytoskeleton. This protein has a known molecular mass of approximately 70 kDa. Within human tissues PLS1 is mainly expressed in the small intestine and inner ear supporting its roles in processes like cellular movement and structural integrity.
