PLSCR3 also known as phospholipid scramblase 3 plays a role in the translocation of phospholipids across cell membranes. It weighs approximately 37 kDa and is closely associated with the mitoplast. This protein is found mainly in the mitochondria heart liver and skeletal muscle tissues. PLSCR3 is vital for maintaining the asymmetry of phospholipid distribution in cellular membranes important for cell viability.
Biological function summary

The activity of PLSCR3 in the mitochondria is significant in apoptosis and lipid metabolism processes. As a part of the scramblase family it collaborates with proteins in the Bcl-2 family to modulate apoptosis. This interaction is important for regulating the release of cytochrome c an important step in the intrinsic pathway of apoptosis. The phospholipid redistribution function of PLSCR3 supports the remodeling of mitochondrial membranes during metabolic processes.

PLSCR3 influences the apoptosis and phospholipid signaling pathways. It interacts with proteins such as Bax and Bak within the apoptosis pathway facilitating the proper execution of cell death during stress responses. In the phospholipid signaling pathway PLSCR3 coordinates with phosphatidylserine and other lipids to regulate cellular signaling processes and membrane dynamics affecting cellular responses to environmental changes.

Mutations or dysregulation of PLSCR3 can contribute to metabolic disorders and certain types of cancer. The protein's role in apoptosis links it to disorders like cardiomyopathy where regulated cell death is important for heart muscle maintenance. Additionally in cancer abnormal PLSCR3 activity impacts cell growth and survival connecting it to oncogenes like MYC which can drive uncontrolled cell proliferation.
