The protein phosphatase methylesterase 1 (PME-1) also known as protein phosphatase 2A methylesterase is an enzyme that regulates protein function by demethylation. PME-1 has a mass of approximately 42 kDa. It primarily resides in the cytoplasm and nucleus of cells with high expression levels in brain tissue. Its function involves the removal of methyl groups from the carboxyl-terminal leucine residue of the catalytic subunit of protein phosphatase 2A (PP2A). This action regulates the activity states of PP2A a major serine/threonine phosphatase.
Biological function summary

PME-1 influences the stability and activity of PP2A a core component of cellular signal transduction pathways. PME-1 forms part of the regulatory complexes of PP2A modulating its methylation state and associated functional properties. This regulation by PME-1 impacts cell cycle control apoptosis and other critical cellular processes. Additionally PME-1 activity affects signaling pathways by altering PP2A's interaction with various substrates and regulatory proteins.

Pathways

The role of PME-1 in cellular mechanisms links it to the Wnt signaling pathway and MAP kinase signaling cascade. PME-1 catalyzes demethylation reactions that impact PP2A's ability to associate with specific regulatory subunits. Through the Wnt pathway PME-1 affects cellular processes such as proliferation and differentiation by regulating beta-catenin activity. PME-1 has associations with cancer and neurodegenerative conditions. In cancer altered PME-1 expression or activity can lead to uncontrolled cell growth and survival acting through PP2A-related pathways. In neurodegenerative diseases such as Alzheimer's PME-1 may affect tau protein phosphorylation by modulating PP2A activity potentially contributing to pathogenesis. As PP2A regulates tau phosphorylation disruptions in its demethylation by PME-1 has been correlated with disease progression.

Demethylates proteins that have been reversibly carboxymethylated. Demethylates PPP2CB (in vitro) and PPP2CA. Binding to PPP2CA displaces the manganese ion and inactivates the enzyme.
