PMF-1 also recognized as Polyamine-modulated factor 1 is a protein with a molecular mass of around 63 kDa. It plays a mechanical role in transcriptional regulation by interacting with other proteins and DNA sequences. PMF-1 is expressed mainly in the nucleus of cells across various tissues. It partners with other transcription factors modulating gene expression essential for cellular growth and differentiation. This interaction underlines its importance in transcriptional control.
PMF-1 engages in modifying transcriptional responses by forming part of the TCF/LEF transcription factor complex. This complex influences the expression of genes involved in cellular proliferation and differentiation. PMF-1 functions by binding to specific DNA sequences enhancing or repressing the transcription of target genes. Through these actions it serves as a regulatory hub for controlling numerous cellular responses including but not limited to cell cycle progression and apoptosis.
PMF-1 interacts significantly within the Wnt signaling pathway. This pathway is pivotal for regulating cellular processes related to development and growth. PMF-1 associates with proteins such as β-catenin facilitating the transcriptional activation of Wnt target genes. Through this pathway PMF-1 helps transmit signals that dictate cellular fate decisions illustrating its importance in maintaining physiological balance.
PMF-1 has notable implications in cancer and neurodegenerative conditions. In cancer its association with the Wnt signaling pathway implies roles in cell proliferation and tumor progression. The interaction with β-catenin further solidifies its connection to oncogenic processes. In neurodegenerative disorders altered PMF-1 activity can influence neuronal growth and survival implicating it in pathologies like Alzheimer's disease through dysregulated transcriptional responses.

PMF-1 was immunoprecipitated from 1mg of HeLa (Human epithelial cells from cervix adenocarcinoma) whole cell extract with Anti-PML Protein antibody [EPR16792] ab179466 at 1/80 (or 5 μg). Western blot was performed using Anti-PMF-1 antibody [EPR17298] ab199531 at 1/1000. Anti-Rabbit IgG (HRP), specific to the non-reduced form of IgG, was used as secondary antibody at 1/1500 dilution.
