The Polyribonucleotide 5'-hydroxyl-kinase 3'-phosphatase often known as PNK or PNKP protein is a dual-function enzyme necessary in DNA repair processes. PNKP with a mass of about 57 kDa performs both kinase and phosphatase activities. It removes 3'-phosphates and phosphorylates 5'-hydroxyl groups in polynucleotides preparing them for subsequent repair. This enzyme is expressed in various tissues with significant levels in the brain indicating its widespread importance in cellular processes.
Biological function summary

PNKP protein is integral to maintaining genome stability by facilitating the repair of single-strand breaks and double-strand breaks in DNA. It is not part of a complex but often collaborates with other repair proteins through its enzymatic functions. PNKP ensures repaired DNA ends are properly processed to allow accurate ligation by DNA ligases contributing to the integrity of the genetic material in cells.

Pathways

PNKP operates mainly in the DNA repair pathways specifically in Base Excision Repair (BER) and Non-Homologous End Joining (NHEJ). Within the BER pathway PNKP collaborates with proteins like DNA polymerase β and XRCC1 in processing damaged DNA. In the NHEJ pathway it associates with proteins like Ku and DNA-PKcs aligning for intricate repair of DNA double-strand breaks.

Mutations and malfunctions in PNKP are linked to neurological disorders and cancer. Dysfunctions in PNKP activity associate with neurodegenerative conditions like Microcephaly with Seizures due to its role in efficient DNA repair in neuronal tissues. Additionally its connection to DNA Repair pathways links PNKP malfunction with increased cancer risk. PNKP involvement with proteins like ATM and ATR signals its importance as these are key players in the cellular response to DNA damage particularly in preventing tumorigenesis.
