PNLDC1 also referred to as PARN-like domain-containing 1 is a protein known for its role as a poly(A)-specific ribonuclease. It functions by trimming the poly(A) tails at the 3' end of RNA molecules which affects RNA stability and translation. PNLDC1 has an approximate mass of 61 kDa. Researchers have detected its expression predominantly in the testes indicating a role in germ cell development. This localization suggests PNLDC1's specific involvement in reproductive biology which marks its distinct function from its homologs.
Biological function summary

The activity of maintaining RNA stability places PNLDC1 as part of an exosome complex similar to the one where its related protein PARN functions. PNLDC1 likely contributes to spermatogenesis through its action in RNA processing. By trimming RNA tails it influences the maturation and maintenance of sperm cells highlighting its significance in reproduction. The protein's activity in RNA metabolism emphasizes its regulatory function in controlling gene expression at the RNA level.

Pathways

The significance of PNLDC1 in regulating RNA stability integrates it into essential biological processes such as the RNA decay pathway. The protein works alongside enzymes like Dicer which handle post-transcriptional control of RNA molecules synergizing to maintain RNA integrity. Within the RNA stability pathway its function intersects with RNA interference pathways sharing roles in ensuring correct RNA turnover and function within cells.

Researchers have associated PNLDC1 with certain reproductive impairments particularly spermatogenic failure. This failure links to suboptimal RNA processing where PNLDC1 function is important. Decreased activity of PNLDC1 can impact proteins like DAZL which play roles in germ cell development. Additionally disturbances in PNLDC1 function can contribute to testicular dysgenesis pointing to its potential as a marker for reproductive diseases and emphasizing its importance in diagnosing fertility issues.

Target data

3'-exoribonuclease that has a preference for poly(A) tails of mRNAs, thereby efficiently degrading poly(A) tails (PubMed : 27515512). Exonucleolytic degradation of the poly(A) tail is often the first step in the decay of eukaryotic mRNAs and is also used to silence certain maternal mRNAs translationally during oocyte maturation and early embryonic development (PubMed : 27515512). May act as a regulator of multipotency in embryonic stem cells (By similarity). Is a critical factor for proper spermatogenesis, involved in pre-piRNAs processing to generate mature piRNAs (PubMed : 34347949).
