Protein O-glucosyltransferase 1 (POGLUT1) also known as Rumi or KTELC1 catalyzes the addition of O-linked glucose to serine residues within epidermal growth factor-like (EGF) repeats. This enzyme plays an important role in protein glycosylation. POGLUT1 possesses an approximate molecular mass of 47 kDa. It predominantly exists in the endoplasmic reticulum of cells. Expression of POGLUT1 occurs in various tissues with notable levels in the heart placenta and skeletal muscle.
Biological function summary

This enzyme modifies certain proteins through glycosylation a process vital for normal protein folding and function. POGLUT1 acts independently rather than as part of a larger protein complex. The glycosylation by POGLUT1 affects cell-cell signaling which is essential in developing tissues. It modifies proteins that control cellular signaling pathways providing structural stability and facilitating correct protein interactions.

Pathways

POGLUT1 functions within the Notch signaling pathway. This pathway controls numerous cell differentiation processes impacting development and cell fate decisions. POGLUT1's glycosylation activity enhances Notch receptor maturation influencing its binding affinity and signaling. Perturbations in this pathway can also involve related proteins such as Fringe proteins which adjust Notch signaling through additional glycosylation mechanisms.

Mutations or dysregulation of POGLUT1 link to Dowling-Degos disease a skin condition characterized by reticulate pigmentation. Alterations in POGLUT1 activity may disrupt normal Notch signaling leading to abnormal skin cell differentiation and proliferation. Additionally changes in POGLUT1 function connect to congenital heart defects associating with misregulated Notch signaling and downstream effects on cardiac development.
