Biological function summary
POLG2 facilitates mtDNA replication by stabilizing the catalytic interaction in the polymerase complex. This complex includes POLG2 and the main polymerase subunit POLG. POLG2 increases the binding affinity of POLG to DNA ensuring efficient and accurate DNA synthesis. The proper functioning of POLG2 in assembling the polymerase complex is necessary for the replication of mtDNA and overall mitochondrial function which directly impacts cell energy production and metabolism.
Pathways
POLG2's activity is integral to the mitochondrial biogenesis and DNA replication pathways. Within these pathways POLG2 works closely with the POLG protein as well as other replication machinery proteins such as TWINKLE helicase. These interactions ensure replication precision and efficiency important for maintaining cellular homeostasis and metabolism. Dysfunction can interrupt the function of these critical pathways leading to reduced mitochondrial output and energy disturbances.
