POLR1C also known as RNA polymerase I and III subunit C functions as a component of RNA polymerases I and III essential for the synthesis of ribosomal RNA precursors and small RNAs. This protein weighs approximately 34.5 kDa and is found ubiquitously expressed in human tissues reflecting its fundamental role in cellular transcription machinery. POLR1C is a subunit important for these polymerases' proper assembly and function.
Biological function summary

POLR1C participates in the formation of the RNA polymerase I and III complexes which are responsible for transcribing ribosomal RNA and small RNAs respectively. These processes are important for protein synthesis and cell growth. As part of these complexes POLR1C interacts with multiple other subunits to ensure the correct transcription of ribosomal RNA which forms the structural and functional core of ribosomes essential for translating mRNA into proteins.

Pathways

POLR1C is integral to the transcription pathways specifically those conducted by RNA polymerases I and III. These pathways play an important role in the synthesis of ribosomal RNA and small RNAs which are critical for protein biosynthesis. In these processes the protein interacts with other components like RPA proteins as well as RPC5 highlighting its involvement in the transcription cycle critical for cellular function and maintenance.

POLR1C mutations have connections to Treacher Collins syndrome and hypomyelinating leukodystrophy. In Treacher Collins syndrome POLR1C interacts with proteins involved in ribosomal biogenesis disrupting this process and causing craniofacial development issues. Similarly in hypomyelinating leukodystrophy aberrations in ribosomal RNA synthesis contribute to neurological symptoms further implicating POLR1C’s pivotal role in transcription machinery and development.

DNA-dependent RNA polymerase catalyzes the transcription of DNA into RNA using the four ribonucleoside triphosphates as substrates. Common component of RNA polymerases I and III which synthesize ribosomal RNA precursors and short non-coding RNAs including 5S rRNA, snRNAs, tRNAs and miRNAs, respectively. POLR1C/RPAC1 is part of the polymerase core and may function as a clamp element that moves to open and close the cleft.
