Supplementary information

PPP1R3B also known as Protein Phosphatase 1 Regulatory Subunit 3B plays a mechanical role in glycogen metabolism. The protein affects the activity of glycogen synthase and glycogen phosphorylase by regulating the substrate specificity of Protein Phosphatase 1 (PP1) its enzymatic partner. PPP1R3B has a mass of approximately 33 kDa and its expression is primarily noted in liver and skeletal muscle tissues. The localization of PPP1R3B to these tissues suggests a specialized role in energy storage and regulation.
Biological function summary

Protein Phosphatase 1 Regulatory Subunit 3B influences glycogen synthesis and degradation. As a part of the PP1 holoenzyme complex PPP1R3B modulates the dephosphorylation of target proteins which controls glycogen metabolism. This regulation enables cells to respond to changes in energy demands effectively. PPP1R3B's interaction with other subunits in the PP1 complex allows for dynamic control of the metabolic processes impacting overall energy homeostasis.

Pathways

Regulation of glycogen metabolism involves several important steps where PPP1R3B integrates into the glycogen biosynthetic pathway and the insulin signaling pathway. By forming associations with other proteins such as glycogen synthase it exerts influence on how glycogen is synthesized and mobilized. In response to signals from upstream kinases such as Akt PPP1R3B works with PP1 to ensure the precise modulation of phosphorylation states that dictate glycogen synthase activity.

Irregularities in PPP1R3B functionality connect to conditions like type 2 diabetes and glycogen storage disease. Disruptions in its normal operation can result in improper glycogen mobilization and storage influencing blood glucose levels. The protein interacts with other regulatory proteins like the PP1 complex and AMPK which are involved in pathways linked to insulin sensitivity and energy balance. Understanding PPP1R3B's role in these disorders could provide insights for therapeutic strategies to manage metabolic diseases.

Target data

Acts as a glycogen-targeting subunit for phosphatase PP1. Facilitates interaction of the PP1 with enzymes of the glycogen metabolism and regulates its activity. Suppresses the rate at which PP1 dephosphorylates (inactivates) glycogen phosphorylase and enhances the rate at which it activates glycogen synthase and therefore limits glycogen breakdown. Its activity is inhibited by PYGL, resulting in inhibition of the glycogen synthase and glycogen phosphorylase phosphatase activities of PP1. Dramatically increases basal and insulin-stimulated glycogen synthesis upon overexpression in hepatocytes (By similarity).
