Immunocytochemistry/ Immunofluorescence - Anti-PPP2R2A Antibody [RP23040118] (AB313413)
Immunofluorescent analysis of HeLa cells fixed with 4% formaldehyde reconstituted in 1X PBS for 10 min at room temperature and permeabilized with 0.1 % Triton X-100 in PBS for 15 min at room temperature labeling endogenous PPP2R2A (PP2AB) with ab313413 at 5 µg/mL concentration followed by Goat anti-Rabbit IgG (H+L) Secondary Antibody, Alexa Fluor® 488 conjugate at 1/2000 dilution. Panel a) shows representative cells that were stained for detection and localization of PPP2R2A protein (green), Panel b) is stained for nuclei (blue) using DAPI. Panel c) is a composite image of Panels a and b clearly demonstrating centromere localization of PP2AB. The images were captured at 60X magnification.

What are recombinant multiclonals?
Recombinant multiclonals are a mixture of recombinant antibodies co-expressed from a library of heavy and light chains. They offer several advantages including:
- - The sensitivity of polyclonal antibodies by recognising multiple epitopes
- - High batch-to-batch consistency and reproducibility
- - Improved sensitivity and specificity
- - Long-term security of supply
- - Animal-free batch production

Biological function summary
PPP2R2A influences several cellular processes through its integration into the PP2A holoenzyme complex. It directs the dephosphorylation of proteins like MAP kinases which are important for signal transduction. By associating with the catalytic and scaffolding subunits of PP2A PPP2R2A modulates cell cycle progression apoptosis and DNA replication. Its involvement in these processes highlights its importance in maintaining cellular homeostasis and function.
Pathways
PPP2R2A is integral to the MAPK/ERK and PI3K/AKT signaling pathways. It regulates the activity of several kinases and phosphatases in these pathways influencing cell growth and survival. In the MAPK/ERK pathway PPP2R2A interacts with proteins like MEK and ERK while in the PI3K/AKT pathway it can connect with the phosphatase PTEN highlighting its regulatory roles in critical cellular signaling networks.
