The protein phosphatase 2 regulatory subunit B also known as PPP2R2B or B beta regulatory subunit plays an important role in cellular processes. It has a molecular mass of approximately 55 kDa. This subunit is part of the protein phosphatase 2 (PP2A) complex and is expressed widely but found at high levels in the brain. In particular its expression is prominent in specific regions such as the cerebellum.
Biological function summary

PPP2R2B acts as a regulatory component of the PP2A holoenzyme complex. PP2A is a serine/threonine phosphatase involved in the negative control of cell growth and division. PPP2R2B is one of the regulatory subunits that directs the PP2A core enzyme to certain substrates influencing dephosphorylation activities. It participates in maintaining normal cellular functions especially within the nervous system by modulating the phosphorylation state of various proteins.

Pathways

PPP2R2B impacts the cell cycle and PI3K/AKT signaling pathways. It negatively regulates these pathways by dephosphorylating target proteins acting as a tumor suppressor. The regulation by PPP2R2B of the PI3K/AKT pathway links it to controlling cell proliferation and survival interacting with proteins like AKT1 and mTOR. The involvement of PPP2R2B in these pathways reflects its pivotal role in ensuring cellular homeostasis and response to extracellular signals.

PPP2R2B is associated with neurodegenerative conditions including spinocerebellar ataxia type 12 and Huntington’s disease. Abnormalities in PPP2R2B function or expression can disrupt normal neuronal activities leading to these disorders. In Huntington’s disease alterations in PPP2R2B contribute to the pathology by affecting the dephosphorylation dynamics of proteins such as huntingtin exacerbating disease symptoms and progression. The B regulatory subunit might modulate substrate selectivity and catalytic activity, and might also direct the localization of the catalytic enzyme to a particular subcellular compartment. Within the PP2A holoenzyme complex, isoform 2 is required to promote proapoptotic activity (By similarity). Isoform 2 regulates neuronal survival through the mitochondrial fission and fusion balance (By similarity).
