Supplementary information

Protein phosphatase 2 regulatory subunit B gamma also known as PPP2R2C or B55-gamma is a regulatory subunit of protein phosphatase 2A (PP2A). It exhibits a molecular mass of approximately 55 kDa. PPP2R2C is expressed in various tissues with higher levels found in the brain and testis. By associating with the core PP2A enzyme it modulates substrate specificity catalytic activity and subcellular localization playing a significant role in PP2A function.
Biological function summary

The protein serves as a regulatory component of the PP2A holoenzyme which consists of a structural A subunit catalytic C subunit and regulatory B subunit like PPP2R2C. This complex performs dephosphorylation functions essential for cell cycle regulation and signal transduction. By coupling with PP2A PPP2R2C influences key cellular processes such as cell growth and differentiation by modulating phosphorylation states of critical proteins.

Pathways

This regulatory subunit PPP2R2C integrates into the PI3K/AKT and MAPK signaling pathways. It serves as a modulatory bridge affecting specific protein interactions and phosphorylation events. Through these pathways PPP2R2C connects with proteins like AKT and ERK coordinating signals that regulate cell survival and proliferation while maintaining balanced phosphatase activity.

PPP2R2C associates with neurodegenerative disorders and certain cancers. Aberrations in PP2A activity regulated by PPP2R2C can lead to Alzheimer's disease where abnormal phosphorylation plays a role in tau protein aggregation. In oncology disruptions in PPP2R2C function may contribute to glioblastoma progression due to altered signaling pathways. These conditions highlight the connection of PPP2R2C with key proteins involved in disease underpinning its relevance in therapeutic research.
