PPP2R5A also known as B56α is a regulatory subunit of protein phosphatase 2A (PP2A) with a molecular weight of approximately 55 kDa. This protein subunit expresses widely in various tissues indicating its important role in multiple biological processes. B56α mainly localizes in the cytoplasm where it associates with other components of the PP2A holoenzyme to perform its regulatory functions.
B56α plays an important role in modulating PP2A activity by influencing substrate specificity and subcellular localization. It is part of the heterotrimeric PP2A complex which also includes a catalytic and a scaffolding subunit. This complex serves critical functions in cell cycle regulation development and apoptosis. By binding specific substrates B56α mediates processes like dephosphorylation affecting signal transduction pathways.
B56α is involved in the regulation of signaling pathways such as the Akt/mTOR and Wnt signaling pathways. In these pathways it interacts with proteins like Akt and GSK3β modulating their phosphorylation states and activities. By acting within these pathways B56α impacts processes such as cell growth survival and differentiation showcasing its significance in cellular homeostasis.
B56α's dysregulation links to cancer and neurodegenerative diseases. In certain cancers altered PP2A activity can affect tumor suppressor pathways involving proteins like p53 leading to unchecked cell proliferation.
