Biological function summary

The protein phosphatase 3 complex involves PPP3CC which contributes to numerous cellular signaling pathways by regulating intracellular levels of calcium. This regulation impacts synaptic transmission neuronal excitability and immune responses. In the neuronal context it helps modulate pathways associated with long-term potentiation and learning by influencing the recruitment and function of neurotransmitter receptors.

Activity summary

The PPP3CC gene encodes the catalytic subunit of the protein phosphatase 3 also known as calcineurin gamma which weighs approximately 58 kDa. This enzyme acts as a calcium/calmodulin-dependent serine/threonine phosphatase that modulates various cellular functions by dephosphorylating target proteins. It is widely expressed across various tissues but its presence is significant in the brain where it plays an important part in neural signaling and synaptic plasticity.

Pathways

PPP3CC contributes a regulatory role to the calcium signaling pathway and the calcineurin-NFAT pathway. In these pathways it interacts with and dephosphorylates the NFAT transcription factors influencing gene transcription related to immune response and neural activities. Through these interactions it shares functional relationships with other proteins like NFAT and calmodulin which also participate in these pathways.

Associated diseases and disorders

Irregular activity of PPP3CC links to neurological disorders such as Alzheimer’s disease and schizophrenia. Altered calcineurin signaling involving the PPP3CC subunit associates with disrupted synaptic functions and neuronal communication in these conditions. Furthermore in Alzheimer's disease the dysregulation may involve other proteins like tau or amyloid precursor protein influencing disease progression and severity.
