The protein of interest PPWD1 also known as Peptidylprolyl isomerase domain and WD repeat-containing protein 1 is a part of the cyclophilin family. This protein has a molecular mass of approximately 105 kDa. It is expressed in a variety of tissues including liver and brain. Mechanically PPWD1 functions as a peptidylprolyl isomerase a type of enzyme that catalyzes the cis-trans isomerization of proline residues in peptides and proteins. This action assists in protein folding which is a critical process for proper cellular function.
Biological function summary

PPWD1 plays a fundamental role in the splicing of pre-mRNA by being a part of the spliceosome complex. This complex is essential for the accurate removal of introns from pre-mRNA therefore shaping the final mRNA transcript that gets translated into protein. The WD40 repeat in PPWD1 helps in mediating protein-protein interactions which is vital for its function in the spliceosome complex. This ensures that PPWD1 interacts efficiently with other spliceosomal components facilitating correct splicing.

Pathways

PPWD1 participates in RNA processing pathways particularly focused on mRNA splicing. It acts in the spliceosomal complex with proteins like snRNPs and various other splicing factors regulating the splicing events. PPWD1 is also linked to pathways governing cellular responses to signals due to its role in managing the diversity of protein products derived from alternative splicing events. This protein influences the pathway in coordination with others such as SFRS1 affecting how cells adapt to signaling changes.

PPWD1 has associations with cancer and neurodegenerative diseases. It relates to cancer due to its role in alternative splicing which can affect oncogene expression and tumor suppressor genes promoting disease development. In neurodegenerative disorders PPWD1's malfunction may impact neuron-specific gene splicing linking it to diseases like Alzheimer's.
