Dipeptidase that catalyzes the hydrolysis of dipeptides with a prolyl (Xaa-Pro) or hydroxyprolyl residue in the C-terminal position (PubMed:17081196, PubMed:35165443). The preferred dipeptide substrate is Gly-Pro, but other Xaa-Pro dipeptides, such as Ala-Pro, Met-Pro, Phe-Pro, Val-Pro and Leu-Pro, can be cleaved (PubMed:17081196). Plays an important role in collagen metabolism because the high level of iminoacids in collagen (PubMed:2925654).

PRD also known as the podocyte retinoid-dependent domain is a protein of interest in several research areas. This protein has a known mass of approximately 62 kDa. PRD expresses in kidney podocytes specialized cells responsible for filtration in the renal glomerulus. The protein plays an important role mechanically in maintaining podocyte structure and function contributing to the integrity of the glomerular filtration barrier.
PRD interacts with cellular mechanisms critical for podocyte stability. PRD associates with lipid rafts specialized membrane structures suggesting a role in signal transduction. It functions as part of a larger protein complex that includes nephrin and podocin essential components of the slit diaphragm in podocytes. PRD is involved in cytoskeletal dynamics regulating actin filament organization important for maintaining cell shape and adhesion.
Several cellular activities incorporate PRD into their processes. It is closely tied to the PI3K/AKT signaling pathway a critical pathway regulating cell survival and growth. In the slit diaphragm context PRD works alongside nephrin and p130Cas forming a signaling cascade that ensures effective communication between podocytes. These pathways help maintain the health of the glomerular filtration system by responding to mechanical and biochemical stimuli.
The disruption of PRD function links to renal diseases such as nephrotic syndrome and focal segmental glomerulosclerosis (FSGS). Mutations or alterations in PRD expression or interaction can impair the slit diaphragm leading to proteinuria and progressive kidney damage. The protein interacts with nephrin and podocin whose mutations are also implicated in these disorders highlighting its importance in preserving kidney function.
