Supplementary information

PREB also known as prolactin regulatory element-binding protein weighs approximately 41 kDa. It acts as a transcription factor that binds to specific DNA sequences to regulate gene expression. Researchers find it expressed in various tissues including the pituitary gland liver and pancreas. Its activity involves the control of prolactin transcription a hormone important for lactation and possibly other genes with related promoter elements.
Biological function summary

PREB regulates gene expression by interacting with mitogenic signaling pathways. As part of a protein complex it influences cellular proliferation and secretion processes. Its transcriptional regulation mechanism allows cells to respond to physiological changes both at developmental stages and in adult organisms. This regulation plays a role in maintaining normal cellular functions indicating its importance in various biological processes.

Pathways

PREB impacts the MAPK/ERK signaling pathway and the pancreatic secretion pathway. These pathways are significant for cellular growth differentiation and secretion. Furthermore PREB interacts with proteins like prolactin and ERK contributing to the regulation of transcriptional responses necessary for effective signal processing. These interactions highlight the involvement of PREB in essential physiological and cellular processes.

PREB relates to conditions such as breast cancer and type 2 diabetes. Its dysregulation has been associated with abnormal cell proliferation seen in breast cancer where it may interact with proteins like estrogen receptor. Furthermore PREB influences insulin regulation in the pancreas linking it to type 2 diabetes. By modulating gene expression it directly or indirectly contributes to the progression or prevention of these disorders.

Target data

Guanine nucleotide exchange factor (GEF) that regulates the assembly of the coat protein complex II/COPII in endoplasmic reticulum (ER) to Golgi vesicle-mediated transport. Selectively activates SAR1A and SAR1B by promoting the exchange of guanosine diphosphate (GDP) for guanosine triphosphate (GTP) in these small GTPases (PubMed : 32358066). In their activated GTP-bound state, SAR1A and SAR1B insert into the membrane of the endoplasmic reticulum where they recruit the remainder of the coat protein complex II/COPII which is responsible for both the sorting of proteins and the deformation and budding of membranes into vesicles destined to the Golgi (PubMed : 32358066).. Was first identified based on its probable role in the regulation of pituitary gene transcription. Binds to the prolactin gene (PRL) promoter and seems to activate transcription.
