Supplementary information

PREP1 also called PBX/knotted 1 homeobox 1 (PKNOX1) is a transcription factor. It weighs approximately 45 kDa. Expression of PREP1 is mainly found in the nucleus of various cell types particularly in tissues with rapid cell proliferation like embryonic development regions spleen and thymus. PREP1 is part of the homeobox gene family and is involved in regulating genes necessary for cellular differentiation and growth.
Biological function summary

The function of PREP1 involves the regulation of numerous developmental processes. It interacts with proteins to form heterodimeric complexes like the PREP1-PBX complex which modulate the transcription of specific target genes. This regulation is important for proper development during embryogenesis and also contributes to hematopoietic cell differentiation. Through these mechanisms PREP1 influences processes underlying organogenesis and cellular identity.

Pathways

The function of PREP1 integrates into the broader context of key regulatory cascades including the Hox gene network and the insulin-signaling pathway. PREP1 interacts with PBX proteins within these pathways affecting the transcriptional programs they control. In particular by influencing the Hox pathway PREP1 plays a role in the spatial patterning of body structures and anterior-posterior axis formation in the embryo. This regulatory activity highlights its importance in the intricate orchestration of developmental cues.

Mutations or dysregulation of PREP1 impact conditions like cancer and diabetes. Abnormal PREP1 expression has been connected to tumorigenesis in organs where its developmental control is critical. For example studies show altered PREP1 levels in certain leukemias with implications for progression and outcome. In diabetes the interplay between PREP1 and insulin-signaling pathways can affect glucose metabolism and pancreatic function linking PREP1 to metabolic disorders where it may cooperate with proteins like FOXO1 in influencing glucose homeostasis.
