Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-PRG2 antibody [EPR29096-502] (AB322142)
Immunohistochemical analysis of paraffin-embedded Human cerebrum tissue labeling PRG2 with ab322142 at 1/5000 (0.095 ug/ml) dilution, followed by a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection). Negative control : No staining in human cerebrum. The section was incubated with ab322142 for 10 mins at room temperature. The immunostaining was performed on a Leica Biosystems BOND® RX instrument. Counterstained with Hematoxylin. Secondary antibody only control : Secondary antibody is a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection). Heat mediated antigen retrieval was performed with Tris-EDTA buffer (pH 9.0, Epitope Retrieval Solution2) for 20 mins.

Immunohistochemistry analysis of Formalin/PFA-fixed paraffin-embedded sections human bone marrow tissue labelling PRG2 with ab322142 at 5000 dilution (B), CD1777 with ab255296 at 2000 dilution (C) and CD42d/GP5 with ab322703 at 500 dilution (D). Opal Polymer HRP Ms + Rb was used as a secondary antibody, and DAPI was used for a nuclear counter stain. Heat mediated antigen retrieval with TrisEDTA buffer (pH 9.0, epitope retrieval solution2) for 20 mins.
Panel A : anti-PRG2 (green; Opal™520), anti-CD177 (magenta; Opal™690) and anti-CD42d/GP5 (gray; Opal™570) on human bond marrow.
Panel B : anti-PRG2 staining basophils in human bone marrow.
Panel C : anti-CD177 staining neutrophils in human bone marrow.
Panel D : anti-CD42d/PGP5 staining megakaryocytes in human bone marrow.
Nuclear DNA was labeled with DAPI (shown in blue).
The section was incubated in three rounds of staining : in the order of ab322142, ab255296 and ab322703 for 30 mins at room temperature. Each round was followed by a separate fluorescent tyramide signal amplification system.
The immunostaining was performed on a Leica Biosystems BOND® RX instrument with an Opal™ 4-color kit. Image acquisition was performed with Leica SP8 confocal microscope.

Biological function summary
PRG2 interacts with other components of the immune system to help mediate inflammatory responses. It is a part of the innate immune response and acts by binding to and neutralizing negatively charged surfaces on cells pathogens and tissues. PRG2 can cause tissue damage by its cytotoxic action when released in large amounts impacting cells within its local environment. This protein does not belong to any known larger protein complex but its activity can influence many immune pathways independently.
Pathways
PRG2 contributes significantly to the immune system's function and the body's defense mechanism against parasites and allergens. The protein plays an important role in the allergic inflammatory pathway where it can interact with interleukins such as IL-5 which influences eosinophil activation and survival. Additionally PRG2 impacts the Th2 cytokine pathway coordinating with proteins like IL-4 to modulate immune responses.
