Supplementary information

PRG3 also known as Proteoglycan 3 or pro-eosinophil major basic protein 2 (ProMBP2) is a protein involved in various mechanical functions in the body. It carries a mass of approximately 22 kDa. The protein shows expression primarily in eosinophils a type of white blood cell and also appears in tissues where eosinophils are present like the lungs and gastrointestinal tract. The PRG3 protein localizes to eosinophil granules which are important for storing and releasing the granule contents that contribute to inflammation and immune response.
Biological function summary

PRG3 plays a role in the human immune system as it is involved in the body's response to parasitic infections and allergic reactions. PRG3 releases cytotoxic proteins that damage the membranes of pathogens and contribute to inflammation. This activity implicates PRG3 in the regulation of immune responses. PRG3 functions in collaboration with other proteins within the eosinophil working together with proteins like eosinophil peroxidase to enhance its immune activity.

Pathways

PRG3's role is significant within immune-related pathways such as the immune response pathway and the allergic pathway. In the immune response pathway PRG3 interacts with other immune system proteins like IL-5 which regulates eosinophil production and activation. Furthermore PRG3 contributes to the allergic pathway where it acts in unison with proteins like histamine amplifying allergic inflammation.

PRG3 has links to asthma and eosinophilic esophagitis. In asthma PRG3 contributes to inflammation and airway hyperreactivity by interacting with cytokines such as IL-5 and IL-13 which exacerbate the condition. In eosinophilic esophagitis PRG3's presence in eosinophil-rich infiltrates aggravates inflammation causing symptoms related to this disorder. Understanding these links provides insights into potential therapeutic strategies targeting PRG3 in related conditions.

Target data

Possesses similar cytotoxic and cytostimulatory activities to PRG2/MBP. In vitro, stimulates neutrophil superoxide production and IL8 release, and histamine and leukotriene C4 release from basophils.
