Supplementary information

PRMT10 also known as Protein Arginine Methyltransferase 10 participates in the methylation of arginine residues in target proteins. This enzyme functions mainly in cellular locations such as the nucleus and the cytoplasm. Weighing around 64 kDa PRMT10 modifies histones through methylation playing a critical role in transcriptional regulation and other chromatin-related processes. Expression of PRMT10 can be seen across many tissues with higher levels found in actively dividing cells due to its role in cell cycle regulation.
Biological function summary

PRMT10 affects chromatin structure and gene expression by methylating histones and other substrates. This protein often associates with complexes involved in transcriptional repression or activation. It targets histones like H3 and H4 influencing the recruitment of transcriptional machinery and chromatin remodelers. Through its enzymatic activity PRMT10 impacts processes like gene silencing DNA repair and signal transduction.

Pathways

The enzymatic activity of PRMT10 integrates with several cellular pathways. It prominently operates in the regulation of the Wnt signaling pathway and the cell cycle control pathway. PRMT10 works alongside related proteins such as β-catenin in Wnt signaling modulating gene expression patterns critical for cell growth and differentiation. Furthermore its interaction with cyclin-dependent kinases (CDKs) highlights its involvement in cell cycle checkpoints.

PRMT10 has been linked to cancer and neurodegenerative diseases. Aberrant expression and function of PRMT10 contribute to tumorigenesis by influencing oncogene and tumor suppressor gene expression. In cancer PRMT10 interacts with proteins like Myc enhancing its transcriptional activity which promotes cell proliferation. In neurodegenerative disorders altered PRMT10 activity affects neuronal gene expression highlighting its potential as a therapeutic target.

Target data

Arginine methyltransferase that can both catalyze the formation of omega-N monomethylarginine (MMA) and symmetrical dimethylarginine (sDMA). Specifically mediates the symmetrical dimethylation of SF3B2. Involved in the regulation of alternative splicing of pre-mRNA (PubMed : 25737013, PubMed : 25979344).
