Pro-MCH also known as pro-melanin-concentrating hormone is a precursor polypeptide that undergoes post-translational processing to produce mature melanin-concentrating hormone (MCH). The precursor weighs approximately 18 kDa before processing. Pro-MCH is expressed mainly in the hypothalamus but can also be found in other brain regions involved in regulating appetite and energy homeostasis.
Biological function summary

The pro-MCH precursor undergoes cleavage to generate MCH which plays a significant role in energy balance and feeding behavior. MCH functions are often linked to neuronal populations in the hypothalamus that regulate hunger signals and it is involved in forming neuronal circuits that influence behavior related to food intake. Although pro-MCH itself is not part of a larger complex the derived MCH interacts with its receptors to transmit signals.

Pathways

Melanin-concentrating hormone interacts with energy homeostasis and reward circuits in the brain. It is engaged in the melanocortin pathway which balances energy intake and expenditure. MCH modulates signaling by interacting with receptors such as MCHR1 and MCHR2 influencing food-seeking behavior and energy regulation processes. Additionally its regulation aligns with orexigenic (appetite-increasing) peptides such as neuropeptide Y (NPY) coordinating energy-related responses.

Aberrant MCH signaling relates to metabolic disorders such as obesity and altered response to stress. Individuals with irregular MCH signaling may exhibit increased appetite and weight gain contributing to metabolic syndrome. The interaction with MCHR1 is significant here as alterations in this receptor are associated with such metabolic changes. Furthermore pro-MCH and its derivative peptides have also been linked to mood disorders with disrupted MCH pathways influencing depression-related behaviors through interactions with proteins like corticotropin-releasing hormone (CRH).
