It operates as part of the proline utilization pathway carrying out key reactions necessary for maintaining amino acid balance and energy homeostasis. Although PRODH functions independently it collaborates with other enzymes in the mitochondrial matrix to facilitate efficient energy production and metabolic regulation.

PRODH is involved in proline and arginine metabolism which intersects with the urea cycle and glutamate metabolism. Its activity impinges on the regulation of key pathways like the Krebs cycle through its product pyrroline-5-carboxylate which further converts to glutamate. In this context PRODH is functionally related to other enzymes such as pyrroline-5-carboxylate reductase which modulates the flow and conversion of metabolites within these pathways.
