Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Profilin 2 antibody - C-terminal (AB191054)
Immunohistochemical analysis of paraffin-embedded human thyroid cancer tissue staining Profilin 2 with ab191054 at 2μg/ml. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 2 μg/ml rabbit anti-PFN2 Antibody, ab191054, overnight at 4°C. Peroxidase Conjugated Goat Anti-rabbit IgG was used as secondary antibody and incubated for 30 minutes at 37°C. The tissue section was developed using HRP Conjugated Rabbit IgG Super Vision Assay Kit with DAB as the chromogen

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-Profilin 2 antibody - C-terminal (AB191054)
Immunohistochemical analysis of paraffin-embedded human thyroid cancer tissue staining Profilin 2 with ab191054 at 5 μg/ml. Heat mediated antigen retrieval was performed in EDTA buffer (pH 8.0, epitope retrieval solution). The tissue section was blocked with 10% goat serum. The tissue section was then incubated with 5 μg/mL rabbit anti-PFN2 Antibody ab191054, overnight at 4°C. Cy3 Conjugated Goat Anti-Rabbit IgG was used as secondary antibody at 1 : 500 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

Biological function summary
Profilin 2 regulates actin dynamics essential for cellular processes like membrane trafficking and signal transduction. It is not typically known to be part of an extensive protein complex but interacts with other proteins including members of the Arp2/3 complex to promote actin filament nucleation and branching. This regulation impacts processes like vesicle transport particularly in neuronal cells where Profilin 2 influences synaptic function and plasticity.
Pathways
Actin dynamics controlled by Profilin 2 play significant roles in both the Rho GTPase and PI3K/AKT signaling pathways. In these pathways Profilin 2 interfaces with Rho-related proteins to modulate the actin cytoskeleton impacting cell adhesion and movement. Its interaction with PI3K elements contributes to cell growth and survival signals aligning Profilin 2's functions with cellular responses to environmental cues.
