Progranulin also known as Granulin or Proepithelin is a secreted glycoprotein with a molecular mass of approximately 68 kDa. It is expressed in a variety of tissues including the brain skin immune cells and certain epithelial cells. The protein consists of multiple cysteine-rich granulin modules which are important for its function. Progranulin plays a mechanical role in cell cycle regulation and wound healing by influencing cell proliferation and migration.
Biological function summary

This protein influences numerous cellular activities. Progranulin acts as a growth factor promoting cell survival and tissue repair. It is also a part of the lysosomal function where it helps to regulate lysosomal dynamics and turnover. It interacts with the receptor sortilin which mediates its uptake and function underlining its importance in maintaining cellular homeostasis.

Pathways

Progranulin participates in the tumor necrosis factor (TNF) pathway and the lysosome acidification pathway. It modulates TNF signaling by interacting with TNF receptors potentially balancing inflammatory responses. In the context of lysosomal pathways progranulin works closely with proteins like cathepsin D to facilitate lysosomal enzyme activity and stability ensuring effective cellular turnover and waste management.

This protein has significant implications in neurodegenerative disorders and cancer. Mutations or alterations in progranulin levels associate with frontotemporal lobar degeneration (FTLD) a form of dementia. Reduced progranulin levels contribute to neuroinflammation and neuronal loss in FTLD. Target data

The protein expressed by the GRN gene is a secreted regulator of lysosomal function and a growth factor involved in processes such as inflammation, wound healing, and cell proliferation. It regulates protein trafficking to lysosomes and lysosomal enzyme activity, facilitating lysosomal acidification which leads to the degradation of mature CTSD by CTSB. It also promotes epithelial cell proliferation by blocking TNF-mediated neutrophil activation, thus preventing the release of oxidants and proteases. Additionally, it modulates inflammation in neurons by preserving neuron survival, axonal outgrowth, and neuronal integrity. Granulin-3, -4, and -7 have distinct roles : Granulin-4 promotes proliferation of epithelial cell line A431 in culture; Granulin-3 inhibits epithelial cell proliferation and induces IL-8 secretion; and Granulin-7 stabilizes CTSD by maintaining its aspartic-type peptidase activity through interaction.
