Flow cytometric analysis of 4% paraformaldehyde fixed 90% methanol permeabilized Jurkat (Human T cell leukemia T lymphocyte) treated with UV-C 50 J/m2, then recovery in serum-free medium for 30h (Right) / Untreated control (Left) labelling Properdin/PFC with ab252531 at 1/50 dilution (1μg). Goat anti-Mouse IgG (Alexa Fluor® 488, ab150113) at 1/2000 dilution was used as the secondary antibody.

Properdin also known as factor P or complement factor P plays an essential role in the immune system. It is a serum protein with a molecular mass of approximately 53 kDa. Properdin is most commonly expressed in monocytes neutrophils and activated macrophages. It serves as a positive regulator in the alternative pathway of the complement system providing stability to the complement convertase complexes C3bBb and C3bBbC3b.
Biological function summary

Properdin enhances the activity of the complement system by stabilizing the C3 and C5 convertase enzyme complexes. These complexes are critical in the amplification of the complement cascade leading to pathogen opsonization and lysis. Properdin does not form part of a complex alone but helps existing complement complexes carry out their functions more effectively. By supporting these complexes properdin contributes to immune surveillance and the host's response to microbial invaders.

Pathways

Properdin operates within the alternative complement pathway and plays a pivotal role in immune defense. It interacts with other components such as C3 and C5 contributing to the pathway's ability to clear pathogens and damaged cells. Properdin's activity also indirectly influences other related proteins in the complement system supporting a coordinated immune response that involves factors beyond the alternative pathway itself.

Properdin deficiencies or dysregulations can lead to increased susceptibility to infections particularly by Neisseria species accounting for recurrent meningococcal disease. Additionally abnormal properdin function links to autoimmune conditions such as systemic lupus erythematosus. This connection is due to its relationship with various complement factors such as C3 which are vital for maintaining immune homeostasis.
