Product details

ab240968 is the carrier-free version of ab190791.

Patented technology
Our RabMAb® technology is a patented hybridoma-based technology for making rabbit monoclonal antibodies. For details on our patents, please refer to RabMAb® patents.

What are the advantages of a recombinant monoclonal antibody?
This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Conjugation ready
Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Compatibility
This product is compatible with the Maxpar® Antibody Labeling Kit from Fluidigm, without the need for antibody preparation. Maxpar® is a trademark of Fluidigm Canada Inc.

Supplementary information

Protamine 2 also known as PRM2 or protamine protein is a nuclear protein with a molecular mass of approximately 14 kDa. It is predominantly expressed in the testes specifically during the spermatogenesis process. Protamine 2 replaces histones in haploid phases of spermatids to ensure the proper condensation of sperm chromatin. Its structure comprises arginine-rich sequences enabling the tight packaging of DNA which is important for mature sperm functionality.
Biological function summary

Protamine 2 plays an essential role in sperm chromatin structure and stability interacting with other nucleoproteins to form a protamine-histone-DNA complex. This interaction is fundamental for the accurate packing of DNA within a sperm cell. Protamine 2 and its partners in the complex are critical for the fertility of males as they impact the genetic information's effective transmission to the next generation. The alteration of Protamine's balance affects the integrity of human sperm leading to reproductive challenges.

Pathways

Protamine 2 integrates into the spermatogenesis pathway coordinating with other proteins like Protamine 1 and transition proteins. Together they conduct transitions from histone-based chromatin to protamine-based packaging. This process is essential within the broader context of the reproductive signaling pathway which includes a range of post-translational modifications central to sperm development and maturation.

Disruptions in Protamine 2 expression link to male infertility as imbalances in Protamine 1 and 2 ratios can result in defective sperm packaging. Evidence also associates Protamine 2 dysregulation with a higher risk of sperm DNA fragmentation which can contribute to disorders such as recurrent pregnancy loss. The protein's performance and interaction with other proteins like histones and nuclear transition proteins point to its importance in maintaining genomic integrity during the paternal genetic material transmission.

Target data

Protamines substitute for histones in the chromatin of sperm during the haploid phase of spermatogenesis. They compact sperm DNA into a highly condensed, stable and inactive complex.
