ab22673 staining human Hela cells by ICC/IF. Cells were formaldehyde fixed, permeabilized in 0.1% Triton and blocked in 3% BSA for 15 minutes at room temperature. The primary antibody was diluted 1/2000 (3% BSA) and incubated with sample for 1 hour. An Alexa Fluor® 488 conjugated goat polyclonal to rabbit, diluted 1/1000 was used as secondary.

Immunofluorescence analysis of HeLa cells, staining Proteasome 20S alpha + beta with ab22673.

Cells were fixed with formaldehyde and permeabilized using 0.1% Triton X-100. Samples were incubated with primary antibody (1/1000) fr 1 hour at room temperature. An AlexaFluor®555-conjugated donkey anti-rabbit IgG was used as the secondary antibody.

The proteasome 20S alpha + beta sometimes referred to simply as the 20S proteasome consists of the catalytic core of the larger 26S proteasome complex. This core degrades ubiquitinated proteins in eukaryotic cells. The 20S proteasome has a barrel-like structure composed of 28 subunits arranged in four stacked rings. Each alpha and beta ring contains seven subunits with masses approximately ranging from 20 to 31 kDa. The proteasome 20S alpha + beta is expressed in the cytoplasm and nucleus of cells important for maintaining protein homeostasis by removing damaged or misfolded proteins as well as short-lived regulatory proteins.
Biological function summary

The proteasome 20S alpha + beta plays a central role in protein degradation ensuring cellular function by regulating the concentration of specific proteins. It is an important component of the ubiquitin-proteasome system (UPS) a complex important for protein turnover. The 20S core particle combines with regulatory particles to form the 26S proteasome which recognizes and unfolds ubiquitinated proteins before translocation into the core for degradation. By controlling the half-life of critical proteins the proteasome influences cellular processes such as the cell cycle apoptosis and immune responses.

Pathways

The ubiquitin-proteasome system including the proteasome 20S alpha + beta is integral to protein homeostasis and the cell cycle control pathway. It removes cyclins key regulators of cell cycle progression illustrating its regulatory function. Another significant pathway is the NF-kB signaling pathway where the proteasome degrades inhibitors to activate NF-kB a transcription factor involved in inflammation and immune responses. These pathways highlight the proteasome's relationship with proteins like cyclins and IkBa emphasizing its role in cellular regulation.

Proteasome dysfunction links to neurodegenerative diseases such as Parkinson's disease and certain cancers. Aberrations in proteasomal activity can result in the accumulation of toxic protein aggregates contributing to neurodegenerative processes. Target data

Component of the 20S core proteasome complex involved in the proteolytic degradation of most intracellular proteins. This complex plays numerous essential roles within the cell by associating with different regulatory particles. Associated with two 19S regulatory particles, forms the 26S proteasome and thus participates in the ATP-dependent degradation of ubiquitinated proteins. The 26S proteasome plays a key role in the maintenance of protein homeostasis by removing misfolded or damaged proteins that could impair cellular functions, and by removing proteins whose functions are no longer required. Associated with the PA200 or PA28, the 20S proteasome mediates ubiquitin-independent protein degradation. This type of proteolysis is required in several pathways including spermatogenesis (20S-PA200 complex) or generation of a subset of MHC class I-presented antigenic peptides (20S-PA28 complex). Inhibits the transactivation function of HIF-1A under both normoxic and hypoxia-mimicking conditions. The interaction with EMAP2 increases the proteasome-mediated HIF-1A degradation under the hypoxic conditions. Plays a role in hepatitis C virus internal ribosome entry site-mediated translation. Mediates nuclear translocation of the androgen receptor (AR) and thereby enhances androgen-mediated transactivation. Promotes MAVS degradation and thereby negatively regulates MAVS-mediated innate immune response.
