Immunocytochemistry/Immunofluorescence analysis of Proteasome 20S beta 6 (green) showing staining in the cytoplasm and nucleus of HeLa (Human epithelial adenocarcinoma cell line) cells (right) compared to a negative control without primary antibody (left). Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with ab3331 in 3% BSA-PBS at a dilution of 1 : 20 and incubated overnight at 4°C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight-conjugated secondary antibody in PBS at room temperature in the dark. F-actin (red) was stained with a fluorescent red phalloidin and nuclei (blue) were stained with Hoechst or DAPI. Images were taken at a magnification of 60x.

Immunofluorescent analysis of Proteasome 20S Y (green) in NIH/3T3 (Mouse embryo fibroblast cell line) cells. The cells were fixed with 4% paraformaldehyde for 15 minutes, permeabilized with 0.1% Triton X-100 in PBS for 15 minutes and blocked with 3% Blocker BSA in PBS for 30 minutes at room temperature. Cells were stained with or without Proteasome 20S Y rabbit polyclonal antibody, at a concentration of 5 μg/mL for 1 hour at room temperature, and then incubated with a Goat anti-Rabbit (H+L) Superclonal Secondary Antibody, Alexa Fluor® 488 conjugate at 1/1000 dilution for 1 hour at room temperature (both panels, green). Nuclei (both panels, blue) were stained with Hoechst 33342 dye. Images were taken at 20X magnification.

Samples were electrophoresed using NuPAGE™ 12% Bis-Tris Protein Gel and the resolved proteins were then transferred onto a Nitrocellulose membrane by iBlot® 2 Dry Blotting System. The blot was probed with the primary antibody (ab3331) and detected by chemiluminescence with secondary antibody using the iBright FL 1000. Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit.

The subunit gets expressed in various cell types throughout the body ensuring that proteins are effectively broken down into peptides. This is essential for maintaining protein homeostasis and recycling faulty proteins.
Biological function summary

The proteasome 20S beta 6 subunit forms part of the 20S core complex an important component of the proteasome. The proteasome operates as the main proteolytic machinery in both nucleus and cytoplasm. The 20S core complex pairs with the regulatory 19S cap to form the 26S proteasome. This assembly degrades ubiquitin-tagged proteins which regulates cell cycle progression apoptosis and DNA repair contributing to cellular function and health.

Pathways

Proteasome 20S beta 6 acts within the ubiquitin-proteasome pathway. This pathway regulates protein turnover affecting critical cellular processes such as cell cycle apoptosis and the immune response. The pathway also links with other proteins such as ubiquitin and E1 E2 and E3 ligases which modify proteins with ubiquitin chains to signal for their degradation.

Proteasome 20S beta 6 may relate to cancer and neurodegenerative diseases like Alzheimer's. The abnormal regulation of the proteasome pathway can lead to the accumulation of undegraded proteins contributing to these diseases. Target data

Component of the 20S core proteasome complex involved in the proteolytic degradation of most intracellular proteins. This complex plays numerous essential roles within the cell by associating with different regulatory particles. Associated with two 19S regulatory particles, forms the 26S proteasome and thus participates in the ATP-dependent degradation of ubiquitinated proteins. The 26S proteasome plays a key role in the maintenance of protein homeostasis by removing misfolded or damaged proteins that could impair cellular functions, and by removing proteins whose functions are no longer required. Associated with the PA200 or PA28, the 20S proteasome mediates ubiquitin-independent protein degradation. This type of proteolysis is required in several pathways including spermatogenesis (20S-PA200 complex) or generation of a subset of MHC class I-presented antigenic peptides (20S-PA28 complex). Within the 20S core complex, PSMB6 displays a peptidylglutamyl-hydrolizing activity also termed postacidic or caspase-like activity, meaning that the peptides bond hydrolysis occurs directly after acidic residues.
