Notes

ab316827 is a BSA and Azide free antibody supplied in an unconjugated
format and it is suitable for sandwich ELISAs to quantify Human Protein Z.
The recommended pair for sandwich ELISA is:

Capture: ab316827, Human Protein Z Capture Antibody (unconjugated)

Detector: Anti-Protein Z antibody [EPR25824-8] - BSA and Azide free (Detector) ab316828, Human Protein Z Detector Antibody (unconjugated)

The reference range value is 31.25-2000 pg/mL.

The recommended antibody orientation is based on internal optimization
for ELISAbased assays. Antibody orientation is assay dependent and
needs to be optimized for each assay type. Please note that the
range provided for this antibody is only an estimation based on the
performance of the product using the recommended antibody pair.
Performance of the antibody pair will depend on the specific
characteristics of your assay. We guarantee the product works in
sandwich ELISA, but we do not guarantee the sensitivity or dynamic
range of the antibody in your assay.

Our carrier-free formats are supplied in a buffer free of BSA,
sodium azide and glycerol for higher conjugation efficiency.
The antibodies are provided at an approximate concentration of
1 mg/ml as measured by the protein A280 method. Use our conjugation
kits for antibody conjugates that are ready-to-use in as
little as 20 minutes with <1 minute hands-on-time and 100% antibody
recovery: available for fluorescent dyes, HRP, biotin and gold.

This product is a recombinant monoclonal antibody, which offers several advantages including:

  • - High batch-to-batch consistency and reproducibility
  • - Improved sensitivity and specificity
  • - Long-term security of supply
  • - Animal-free batch production

For more information, read more on recombinant antibodies.

Our carrier-free antibodies are typically supplied in a PBS-only formulation, purified and free of BSA, sodium azide and glycerol. The carrier-free buffer and high concentration allow for increased conjugation efficiency.

This conjugation-ready format is designed for use with fluorochromes, metal isotopes, oligonucleotides, and enzymes, which makes them ideal for antibody labelling, functional and cell-based assays, flow-based assays (e.g. mass cytometry) and Multiplex Imaging applications.

Use our conjugation kits for antibody conjugates that are ready-to-use in as little as 20 minutes with 1 minute hands-on-time and 100% antibody recovery: available for fluorescent dyes, HRP, biotin and gold.

Activity summary

Protein Z also known as PZ is a vitamin K-dependent plasma protein with a molecular mass of approximately 62 kDa. It is primarily expressed in the liver and circulates in the blood. This protein plays a mechanical role in the coagulation system by interacting with the protein Z-dependent protease inhibitor which downregulates blood coagulation contributing to hemostasis. Protein Z is different from other coagulation proteins due to its lack of enzymatic activity but impacts the regulation of thrombin formation.

Biological function summary

Protein Z serves as a significant factor in the coagulation cascade being a part of the protein C pathway complex. Although it lacks enzymatic action Protein Z interacts with other proteins such as Factor Xa and Protein Z-dependent protease inhibitor to exert its effects. Its involvement helps modulate clot formation and stabilize the coagulation process impacting blood flow regulation.

Pathways

Protein Z is intricately linked to the coagulation and thrombosis pathways. It associates with proteins including Factor Xa and Protein C. Protein Z assists in regulating thrombin generation by inhibiting excess coagulation maintaining a balance within the hemostatic system. This interaction ensures the rapid and effective stopping of blood flow when required while preventing unwarranted thrombosis a critical aspect of circulatory system maintenance.

Associated diseases and disorders

Protein Z levels link to thrombosis and hemorrhagic disorders. Abnormalities in Protein Z levels have connections with deep vein thrombosis due to its role in regulating coagulation balance. The protein Z-dependent protease inhibitor's association highlights its significance in clot formation and thrombosis prevention. Additionally deficiencies in Protein Z have connections with bleeding disorders emphasizing the protein's role in maintaining a balanced coagulation process essential for preventing excessive bleeding or clotting.
