Supplementary information

PRPF3 also known as Pre-mRNA processing factor 3 plays an important role in the spliceosome machinery necessary for the splicing of pre-mRNA. It is part of the U4/U6-U5 tri-snRNP complex which is important for the proper assembly of the spliceosome. This protein weighs approximately 77 kDa and can be found in the nucleus of cells where mRNA splicing takes place. Its expression occurs across various tissues reflecting its relevance in general cellular functions.
Biological function summary

PRPF3 is involved in mRNA processing by facilitating the removal of introns from pre-mRNA transcripts. In its role within the spliceosome it works alongside other proteins in the complex to ensure accurate intron excision and exon ligation. This process is essential for creating mature and functional mRNA which translates into proteins vital for cellular operations. PRPF3 ensures the precision of RNA splicing affecting gene expression regulation and influencing alternative splicing events.

Pathways

PRPF3 is integrated into the spliceosomal cycle and is essential for the regulation of gene expression. This protein interacts with other spliceosome-associated proteins such as PRPF8 and SNRNP200 participating in the assembly and activation of the spliceosome. Its function influences the mRNA surveillance pathway ensuring only properly spliced mRNAs are translated into proteins which is a critical regulatory mechanism in cells.

Mutations in the PRPF3 gene have been linked to Retinitis Pigmentosa 18 a genetic disorder leading to progressive vision loss. The dysfunction can impact the interactions of PRPF3 with other spliceosomal proteins such as PRPF8 disrupting normal RNA splicing and leading to photoreceptor cell death. Additionally aberrant splicing events involving PRPF3 can contribute to other genetic conditions where precise splicing is impaired highlighting its importance in maintaining cellular homeostasis.
