Actin is a highly conserved protein that polymerizes to produce filaments that form cross-linked networks in the cytoplasm of cells (PubMed:29581253). Actin exists in both monomeric (G-actin) and polymeric (F-actin) forms, both forms playing key functions, such as cell motility and contraction (PubMed:29581253). In addition to their role in the cytoplasmic cytoskeleton, G- and F-actin also localize in the nucleus, and regulate gene transcription and motility and repair of damaged DNA (PubMed:29925947).

ab317803 is the carrier-free version of Anti-PSD95 antibody [EPR23124-118] - Synaptic Marker – Goat IgG (Chimeric) ab317793.
This goat monoclonal chimeric antibody has been engineered from a RabMAb parent antibody (Anti-PSD95 antibody [EPR23124-118] - Synaptic Marker ab238135). By necessity, some rabbit sequence is retained as part of the variable domain. When multiplexing with other rabbit-derived antibodies, using cross absorbed Fc-reactive secondary antibodies are recommended.

Immunofluorescence staining of PSD95 in sections of formalin-fixed paraffin-embedded rat brain (positive) and rat liver (negative).
Performed on a Leica BOND. The section was pre-treated using heat mediated antigen retrieval with sodium citrate (pH6.0) for 20 minutes. The section was then incubated at room temperature for 1 hour with ab317803 at 1/100 dilution, and then incubated for 1 hour with Donkey Anti-Goat IgG H&L (Alexa Fluor® 647) preadsorbed ab150135 Donkey Anti-Goat IgG H&L (Alexa Fluor® 647) preadsorbed at 1/1000 dilution (shown in magenta). Nuclear DNA was labelled with DAPI (shown in blue). The section was then mounted using Dako Fluorescence Mounting Medium®.
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
For other IHC staining systems (automated and non-automated), customers should optimize variable parameters such as antigen retrieval conditions, antibody concentrations and incubation times.
