PSMD11 also known as RPN-6 is a regulatory subunit of the proteasome 26S. It is approximately 50 kDa in mass and plays an essential role in the proteasome's function responsible for protein degradation within cells. PSMD11 is broadly expressed in various tissues including high expression in the brain heart and skeletal muscle. Its mechanical role involves binding to other subunits to stabilize the proteasome structure and facilitate substrate entry into the proteolytic core.
Biological function summary

PSMD11 is essential for the proper assembly and activity of the 26S proteasome complex which is important for ubiquitin-mediated proteolysis. This process controls protein turnover and regulates the concentration of various proteins to maintain cellular homeostasis. PSMD11 enhances the degradation of misfolded or damaged proteins therefore preventing the accumulation of toxic protein aggregates within cells. Component of the 26S proteasome, a multiprotein complex involved in the ATP-dependent degradation of ubiquitinated proteins. This complex plays a key role in the maintenance of protein homeostasis by removing misfolded or damaged proteins, which could impair cellular functions, and by removing proteins whose functions are no longer required. Therefore, the proteasome participates in numerous cellular processes, including cell cycle progression, apoptosis, or DNA damage repair. In the complex, PSMD11 is required for proteasome assembly. Plays a key role in increased proteasome activity in embryonic stem cells (ESCs) : its high expression in ESCs promotes enhanced assembly of the 26S proteasome, followed by higher proteasome activity.
