PSMF1 also known as proteasome inhibitor subunit 1 or PI31 inhibits the proteasome a protein complex responsible for degrading unwanted or damaged proteins through ubiquitin-proteasome pathway. PSMF1 has a mass of approximately 29 kDa and shows expression in a variety of human tissues with notable presence in the liver brain and testis. Its role as an inhibitor allows regulation of proteasomal activity playing important part in maintaining protein homeostasis.
PSMF1 interacts with proteasomal subunits functioning within the core particle of the 20S proteasome complex. Its inhibitory action helps balance protein degradation in cells impacting processes such as cell cycle apoptosis and stress responses. The modulating effect of PSMF1 on the proteasome distinguishes its role in cellular regulatory mechanisms.
PSMF1 contributes to the ubiquitin-proteasome pathway and influences the NF-kB signaling pathway. PSMF1's interactions within this system connect it with other essential proteins including ubiquitin which mark proteins for degradation and the proteasome the complex where degradation occurs. This process affects the transcription factor NF-kB which regulates genes involved in immune response and inflammation.
PSMF1 has been associated with cancer and neurodegenerative diseases like Parkinson’s disease. In cancer its role in regulating proteasome activity impacts cellular proliferation and survival connecting it to other proteins such as p53 involved in tumor suppression. In neurodegenerative disorders impaired proteasome function links PSMF1 to the accumulation of misfolded proteins with proteins like alpha-synuclein involved in the pathology of conditions such as Parkinson’s disease.
